Abstract

Duplex DNA is rarely considered today to be a structurally monotonous molecule. Although generally viewed as having an overall conformation very similar to that first proposed by Watson and Crick (1953) and refined by others (Arnott and Hukins, 1972), research over the past several years has clearly demonstrated that duplex DNA is capable of adopting a wide range of conformations which differ from one another in both subtle and dramatic ways (Leslie et al., 1980: reviewed in Wells et al., 1980; Dickerson et al., 1982; Zimmerman, 1982; and Singleton, 1985). Conformational heterogeneity is manifest in both an environmental-dependent and sequence-dependent manner, as illustrated by the existence of DNA structural families — A, B, C, etc. — and geometric variations within a given family. Investigations concerning DNA polymorphism stem not only from a desire to advance the knowledge of DNA structure per se, but also from the firm belief that DNA structural heterogeneity plays a significant role in the biological processes which involve DNA replication, recombination, transcription and gene regulation (Wells et al., 1980).
