X-ray crystallographic studies of nucleic acid binding proteins

Abstract

In recent years several symposia and conferences have focused upon the topic of biological recognition, particuarly as it applies to protein-nucleic acid systems (Eisenberg et al., 1979; Parsegian, 1980; Vogel, 1977; Sarma, 1980, 1981). Attempts have been made to delineate the atomic interactions by which these macromolecules recognise and bind to one another, since such knowledge could clarify some of the mechanisms by which the flow of genetic information is mediated. Sessions dealing directly with the question of those interactions as they exist at an atomic level have, unfortunately, been primarily characterised by educated speculation. Few structures of true protein-nucleic acid complexes had been determined by X-ray diffraction analysis, which is the only method currently available for the direct visualisation of molecular detail.
