Abstract
The similarity between the positive inotropic effects of cardioactive steroids and calcium ions on the heart muscle led Loewi (1918) to suggest that glycosides sensitize the heart to calcium. There is evidence that calcium ions play an important role in activating myofibril-ATPase, so such a sensitization could mean that the cardioactive steroids act directly on the contractile proteins and make them sensitive to calcium ions. Many investigations, however, show that the contractile proteins are not directly affected by the glycosides in a manner which could explain their inotropic action.
