Attempt at Heterologous Expression of Clostridial Hydrogenase in Cyanobacteria

Abstract

A shuttle vector (pKE4-9) system for a cyanobacterium, Synechococcus sp.PCC7942 and Escherichia coli, was examined for expression of clostridial hydrogenase. pKE4-9 carries the chloramphenicol-resistant (cat) gene and a strong promoter we have isolated from PCC7942. The clostridial hydrogenase gene was cloned upstream of the cat gene in pKE4-9, which was transformed into PCC7942. Although the cat protein was expressed, the clostridial hydrogenase was not detected by Western blot analysis. The hydrogenase mRNA was detected in PCC7942; it was co-transcribed with cat. The problems in the expression of foreign proteins in cyanobacteria are discussed.
