Sucralfate enhances ulcer healing and protects the mucosa of the stomach against damage induced by various irritants in human subjects as well as in experimental animals. Since sucralfate does not affect gastric acid secretion, other mechanisms are responsible for its protective and healing effects. Sucralfate’s efficacy in the prevention of gastric mucosal injury induced by various irritants mimics to some extent the protection afforded by prostaglandins. The hypothesis that sucralfate enhances mucosal defense and induces ulcer healing via stimulation of endogenous gastric prostaglandin synthesis was therefore thoroughly investigated in experimental models and also in human subjects. Whereas in many studies the direct effect of sucralfate on gastric generation and release of prostaglan- dins was evaluated, another approach elucidated the effect of cyclooxygenase inhibitors such as indomethacin on the protection afforded by sucralfate.
