La valvola polmonare si trova anteriormente ed a sinistra della valvola aortica. Le tre cuspidi polmonari assumono la forma di una semiluna (semilunari). Queste sono morfologicamente simili ma di dimensioni diverse fra loro. I seni coronarici aortici destro e sinistro sono sempre di fronte alla valvola polmonare. Le cuspidi polmonari sono più sottili e più delicate delle cuspidi aortiche.
