Abstract

The phosphatidylinositol-3-kinase/protein kinase (PI3K/PKB) signaling pathway—also known asthe survival or anti-apoptotic pathway—plays an important role in controlling cell growth, proliferationand survival. Whatever the mechanism, the prevalence of PI3K/PKB signaling abnormalities in human cancercells has suggested the potential use of PI3K/PKB pathway modulators as novel targeted therapeutic agents.Although often non-selective for the intended target, early PI3K/PKB inhibitors have been extensively usedas molecular probes for improving our understanding of the biological processes associated with this pathway.A few of these early compounds, or closely related analogues, have provided proof-of-concept in preclinicalsettings and have entered clinical trials. This work covers recent salient achievements in the identificationand development of PI3K/PKB pathway modulators, updating recent reports on this class of potential targetedanticancer drugs.
