Biocompatibility, Cytotoxicity and Bioactivity of Amorphous Carbon Films

Abstract

In this work we investigate the possibility of using an amorphous carbon layer as a bioactive coating that promotes bone ingrowth for medical implants in direct contact with bone. The initial step was to confirm the tissue compatibility of the amorphous carbon coatings by studying cellular adhesion, proliferation and viability with human osteoblasts and fibroblasts, cultured on the carbon surface. To investigate the bone-bonding capabilities of the sputtered a-C coatings, “in vitro” mineralisation studies were performed. For the biomineralisation assays, human osteoblasts were grown on a-C samples for periods up to 14 days, leading to the formation of mineralised structures that were morphologically and biochemically investigated by the scanning electron microscope and X-ray microanalysis, respectively.
