Immunomodulation by Mycoplasmas: Artifacts, Facts and Active Molecules

Abstract

One of the earliest reports on mycoplasma-mediated immunomodulation was a study concerning the toxicity of heat-killed mycoplasmas28. As is now known, bacterial toxicity, if not caused by exotoxins, is the result of excessive stimulation of the innate immune system through the so-called bacterial modulins; these are mostly components of the cell wall35 and, possibly, pieces of nucleic acids containing the motif CpG51. Cells of the innate immune system, in particular of the monocyte/macrophage lineage, react to these components by release of proinflammatory cytokines and IFN-γ, which in turn can lead to shock and death116. When the observations of Gabridge and Murphy28 were made, knowledge about these potent mediators of various cellular responses was nonexistent and the data were uninterpretable in modern terms. However, their report is a good example of a study which intentionally dealt with mycoplasmas. Many more studies were published, and some until quite recently, in which immunomodulation due to the presence of mycoplasmas was unknowingly observed and reported.
