Mechanism of inhibition of prostaglandin biosynthesis by anti-inflammatory steroids

Anti-inflammatory steroids inhibit prostaglandin (PG) biosynthesis in a variety of tissues and cultured cells. This inhibition is secondary to a suppression of phospholipase A2, the enzyme which releases arachidonic acid from membrane phospholipids1,2. Recent reports show that the inhibition of PG biosynthesis and the anti-inflammatory effect of glucocorticosteroids depend on receptor occupancy and require RNA and protein synthesis3–7.
