Abstract

This is the simplest cloning strategy, which uses two RE sites in the MCS of a vector. Below are the step by step descriptions of the cut and paste method (Fig. 3.1).

    The primer mentioned here has nothing to do with the RT reaction. Strictly speaking, primers bind to the cDNA synthesized from the RT reaction, but they are commonly called RT-PCR primers . To be accurate, they should be called cDNA primers
