Zusammenfassung
Die Methoden für die Identifizierung und Charakterisierung „idealer“ Enzyme für den industriellen Einsatz im Bereich „Weißer“ Biotechnologie haben sich in den letzten Jahren rasant weiterentwickelt. Der Einsatz aktivitätsbasierter Methoden für die Enzymidentifizierung aus natürlichen Ressourcen wie dem Metagenom erlaubt das Auffinden selektiver und aktiver Biokatalysatoren. Der wesentliche Vorteil eines aktivitätsbasierten Screenings ist die direkte Identifizierung gewünschter Enzymaktivitäten völlig unabhängig von jeder Kenntnis über Sequenz oder Struktur eines Enzyms. Auf diese Weise ermöglicht das aktivitätsbasierte Screening auch die Identifizierung neuer, bisher unbekannter Enzyme. In diesem Kapitel werden die Ressourcen für eine Enzymidentifizierung sowie die strategische Herangehensweise in einem Multiparameterscreening beschrieben. Außerdem werden die verschiedenen Methoden, die in einer solchen Screeningkampagne zum Einsatz kommen, deren Vor- und Nachteile und Kombinationsmöglichkeiten erläutert.
