FormalPara Synthese – Verteilung – Abbau – EliminationDas Pharmakon wird oral appliziert und rasch resorbiert. Es wird in der Leber zu phenolischen Verbindungen metabolisiert, die teils konjugiert, teils unkonjugiert renal (etwa 65 %) bzw. enteral (etwa 35 %) eliminiert werden; 1 % der Dosis wird unverändert im Urin ausgeschieden.
FormalPara Halbwertszeit1–2 Stunden (Plasma).
FormalPara Funktion – PathophysiologieDiclofenac hemmt die Cyclooxigenase und damit die Prostaglandinsynthese. Bei kardialer, renaler oder hepatischer Insuffizienz sowie Magen-/Darmulkus sollte Diclofenac nicht appliziert werden. Insbesondere bei gleichzeitiger Therapie mit Antikoagulanzien und Antidiabetika sind Blutgerinnung bzw. Glukosekonzentration im Blut zu überwachen. Die Wirkung von Diuretika und Antihypertonika ist unter Diclofenacbehandlung vermindert.
