FormalPara DefinitionZur Abschätzung des Leber-Eisen-Gehalts durchgeführte, semiquantitativ (färberisch) oder quantitativ (Atomabsorptionsspektrometrie) ermittelte Eisenkonzentration im Leberparenchym (dividiert durch die Zahl der Lebensjahre).
FormalPara BeschreibungEin Leberbiopsiezylinder von etwa 2,5–3 cm Länge wird aliquotiert in Fragmente von 0,5–1,0 cm und entweder sofort tiefgefroren oder in 10 % Formalin fixiert. Der Eisen-Gehalt kann semiquantitativ durch Anfärbung mit Perls Preußischblau (Eisenfärbung mit saurem Ferrocyanid) und nachfolgender Auswertung anhand eines Scoring-Systems (z. B. Ludwig-Batts-System mit Bestimmung des prozentualen Anteils eisengefärbter Hepatozyten) oder quantitativ mittels Atomabsorptionsspektrometrie nach Salpetersäureaufschluss erfolgen.
Normalkonzentrationen:

Männer: ≤30 μmol/g Trockengewicht

Frauen: ≤17 μmol/g Trockengewicht
Konzentrationen >40 μmol/g Trockengewicht weisen auf eine deutliche Eisenüberladung, z. B. bei Hämochromatose hin.
