Unter Verwertung eigener Erfahrungen zunächst am Bakterienenzym [Franke, W., u. GL E. Hahn: H. 299, 15 (1955)]. Die dort erwähnte Zerstörung (Disproportionierung) von Glyoxylat unter den Bedingungen der alkalischen Allantoinhydrolyse ist nach späterer Nachprüfung (Franke, W., u. A. Thiemann: Unveröffentlicht) zu streichen; sie spielt bei der oben angegebenen Methode auch keine Rolle. Die obige Methode hat sich auch beim Soja-Enzym bewährt.
Diese hier und später (S. 381) angegebene Enzymmenge gilt nur für Rohextrakte; von gereinigten Enzymlösungen ist ein kleineres Volumen auf 2 ml zu verdünnen.
Die hier verwendete Allantoin-Gesamtkonzentration von 0,005 m ergibt beim Soja-Enzym rund 3/4 der maximalen Reaktionsgeschwindigkeit; die letztere wird fast erreicht, wenn die 4 ml Wasser des Ansatzes ebenfalls durch Allantoinlösung ersetzt werden.
