Summary
Highly purified mitochondria isolated from a variety of vertebrate tissues have the enzymic apparatus to incorporate radioactively labelled precursors like deoxycytidine monophosphate, thymidine or thymidine triphosphate (TTP) into mitochondrial DNA. The incorporation rate depends on the mitochondrial preparation used: With “intact” mitochondria a comparatively low rate of incorporation is obtained, obviously due to a permeability barrier which prevents the desoxynucleoside triphosphates from penetrating the inner compartment of the cell particles. Mitochondria prepared in sucrose media of high molarity or mitochondrial preparations obtained by breaking their structure with the “Ultraturrax”-homogenizer show a considerable increase in the rate of 3H-TTP-incorporation. In order to obtain a maximal rate all 4 nucleoside triphosphates have to be present in the incubation medium. The incorporation of 3H-TTP is inhibited by typical inhibitors of DNA-polymerase reactions.
Data on some of the enzymic parameters of the mitochondrial DNA-dependent DNA-polymerase in vertebrate tissue are presented such as the dependence of the enzymic reaction on the amount of enzyme, on divalent cations, on addition of DNA as well as a comparison with the corresponding reaction located in the cell nucleus.
The DNA-polymerase activity differs from the corresponding nuclear enzyme in a number of characteristics. The enzyme does not represent a terminal desoxynucleotidyl transferase, like most of the 3H-TTP incorporating activity found in the hyaloplasmic fraction.
Since it has been possible to solubilize the mitochondrial enzyme there is no major difficulty in purification.
So far, a net synthesis of mitochondrial DNA has not been demonstrated but the capacity of the enzyme would allow the conclusion that the enzyme studied accounts for the synthesis of mitochondrial DNA observed in vivo.

In dieser Arbeit berichten wir über Eigenschaften der mitochondrialen DNA-Polymerase und deren Abgrenzung gegenüber entsprechenden Reaktionen des Zellkerns.
Isolierte Mitochondrien der verschiedensten Wirbeltierorgane verfügen über Fermentsysteme, die Deoxynucleotide (radioaktiv markiertes dCMP, Tdr oder TTP) in die Mitochondrien-DNA einbauen können. „Intakte“ Mitochondrien zeigen einen geringeren Einbau als Mitochondrien, deren Membran vorgeschädigt ist (z.B. Behandlung mit dem „Ultraturrax“-Homogenisator oder osmotischer Schock). Für einen maximalen Einbau ist die Anwesenheit aller vier Nucleotide im Versuchsansatz erforderlich. Typische Hemmstoffe von DNA-Polymerasen wie Nogalamycin, Cinerubin A, Acriflavin, Phleomycin u. ä. inhibieren die Reaktion.
Durch verschiedene Parameter — Abhängigkeit von der Fermentmenge, vom pH, von der Deoxynucleotidkonzentration, von der Gegenwart zweiwertiger Kationen, vom Zusatz exogener DNA — wurde das Ferment in Mitochondrien näher charakterisiert. Das untersuchte Ferment gehört nicht zur Gruppe der terminalen Deoxynucleotidyl-Transferasen.
Das mitochondriale Ferment läßt sich deutlich gegen die Zellkern-Polymerase und teilweise von einem entsprechenden Ferment im Hyaloplasma abgrenzen.
Da es möglich ist, das mitochondriale Ferment in Lösung zu bringen, bestehen keine grundsätzlichen Schwierigkeiten, das Enzym zu reinigen und noch eingehender zu untersuchen.
Ob die von uns charakterisierte mitochondriale DNA-Polymerase zur Verdopplung der ringförmigen Mitochondrien-DNA führt, ist zur Zeit nicht zu beurteilen. Nach einer Überschlagsrechnung der Synthesekapazität kommt das Enzym durchaus für eine Replikation der mitochondrialen DNA in Frage.
