Summary
With the help of the histochemical fluorescence method the adrenergic innervation of the frog heart (Rana temporaria) and the uptake of catecholamines by the adrenergic nerves after pretreatment with reserpine was investigated.

The atrium of the frog heart contains numerous adrenergic nerve fibers (axons and terminals with varicosities) which form a network in the sinuauricular node. In the ventricle, the bulk of adrenergic terminals with varicosities was localised near the atrioventricular node.

After pretreatment with reserpine the fluorescence disappeared from the atrium as well as from the ventricle. Administration of adrenaline, noradrenaline and the respective α-methylated derivatives resulted in a restoration of the fluorescence of the adrenergic nerves but without reappearance of the varicosities, so that the nerves appeared to be smooth.
The fluorescence intensity of the adrenergic nerves was high after injection of α-methyladrenaline, somewhat lower after α-methylnoradrenaline and adrenaline and very low after the administration of noradrenaline. Its restoration was time and dose dependent. Maximal fluorescence was reached within 1 hr after the injection of 2 mg/kg α-methyladrenaline. Pretreatment with amphetamine and des-methylimipramine inhibited the uptake.

After incubation of frog atria in vitro a dose dependent uptake of α-methyladrenaline and adrenaline by the adrenergic nerves could be observed. This uptake of catecholamines takes place during incubation at 25° and 0°C.

It is concluded that the adrenergic nerves of the frog heart contained an reserpine-resistant mechanism of uptake for catecholamines which is localised in the cell membrane of the nerves operating at high and low temperatures.
Zusammenfassung
Mit Hilfe der histochemischen Fluorescenzmethode zur Darstellung biogener Monoamine wurden die adrenergische Innervation des Froschherzens (Rana temporaria) und die Aufnahme von Brenzcatechinaminen in die adrenergischen Nerven nach Vorbehandlung mit Reserpin untersucht.

Der Vorhof des Froschherzens enthält zahlreiche adrenergische Nervenfasern (Axone und Nervenendigungen mit kleinen Varicositäten), die im Sinusknoten ein Netzwerk bilden. Im Ventrikel waren zahlreiche varicöse Nervenendigungen sichtbar, besonders im Bereich des atrioventrikulären Knotens.

Nach Vorbehandlung mit Reserpin (3 Ö 25 mg/kg) war in Vorhof und Kammer keine oder nur sehr geringe spezifische Fluorescenz zu beobachten. Injektionen von Adrenalin, Noradrenalin und den entsprechenden α-methylierten Verbindungen restituierten die Fluorescenz der adrenergischen Nerven in Atrium und Ventrikel, wobei sich jedoch die Varicositäten der Nervenendigungen nicht darstellten.
Die Fluorescenzintensität der adrenergischen Axone war am stärksten nach Injektionen von α-Methyladrenalin, weniger stark nach Verabfolgung von α-Methylnoradrenalin und Adrenalin und am schwächsten nach Noradrenalin. Ihre Restitution war zeit- und dosisabhängig; das Maximum der Fluorescenz wurde nach 1 Std mit 2 mg/kg erreicht. Vorbehandlung mit Amphetamin und Desmethylimipramin hemmte die Aufnahme.

Auch in vitro ließ sich eine konzentrationsabhängige, weitgehend temperaturunabhängige (25° und 0°C) Aufnahme von a-Methyladrenalin und Adrenalin nachweisen.

Die Ergebnisse sprechen dafür, daß auch beim Kaltblüter (Frosch) Reserpin die Aufnahme von Brenzcatechinaminen durch die Zellmembran der adrenergischen Nerven nicht beeinträchtigt.
