Zusammenfassung
Zu Beginn des 20. Jahrhunderts begann Archibald E.Garrod in England seine klassischen Untersuchungen über die Alkaptonurie, die man daran erkennt, dass sich der Urin beim Stehen an der Luft allmählich verdunkelt. Garrod untersuchte den Urin der Patienten und konnte dabei eine große Menge Homogentisinsäure nachweisen, die beim Abbau der aromatischen Aminosäuren Phenylalanin und Tyrosin entsteht. Dieses Abbauprodukt ist normalerweise im Urin nur in Spuren nachweisbar, da es in Leber und Nieren vollständig abgebaut werden kann.Garrod stellte außerdem fest, dass diese Störung auch bei Verwandten der Patienten auftrat. Er zog daraus den Schluss, dass diese Krankheit durch den erblichen Defekt eines spezifischen Enzyms zustande kommt. Etwa 50 Jahre später (1958) wurde seine Hypothese bestätigt.Garrod prägte damals den heute noch gebräuchlichen Ausdruck der angeborenen Stoffwechselerkrankungen.
