Bestimmung des Profils inflammatorischer Mediatoren bei Helicobacter pylori-assoziierter Gastritis mittels RT-PCR

Summary

Helicobacter pylori-associated gastritis is characterized by a dense infiltration of inflammatory cells. Because H. pylori is a non-invasive bacteria, a signal transduction has to exist which results in an accumulation of granulocytes and lymphocytes into the lamina propria. Chemokines are such chemotactic mediators. The transcripts of these mediators were analyzed in antral biopsies of patients with H. pylori gastritis by means of reverse transcriptase polymerase chain reaction (RT-PCR). The profile of inflammatory mediators showed that H. pylori infection upregulates interleukin (IL)-8 but not ENA-78. This implies that the recruitment of neutrophils, which is a sign of the activity of gastritis, is stimulated by IL-8. In the chronic phase of gastritis the lymphocyte activating cytokine MIP-1α mainly mediating the signal.

Zusammenfassung

Die Helicobacter pylori-assoziierte Gastritis ist gekennzeichnet durch die Infiltration immunreaktiver Zellen. Da H. pylori nicht invasiv ist, muß eine Signalkette existieren, die chemotaktisch auf die Granulozyten und Lymphozyten wirkt. Chemokine sind z.B. solche signaltransduzierenden Mediatoren, deren Transkripte wir mit Hilfe der RT-PCR Methode in antralen Biopsien von Patienten mit H. pylori-Gastritis nachweisen konnten. Das Profil inflammatorischer Mediatoren erbrachte, daß bei der H. pylori-assoziierten Gastritis eine erhöhte Transkription von IL-8, nicht dagegen von ENA-78 vorliegt. Dies zeigt an, daß für die Akkumulation der Neutrophilen in der aktiven Gastritis IL-8 und nicht ENA-78 verantwortlich ist. Dagegen vermittelt in der chronischen Phase der Gastritis das Lymphozyten aktivierende Zytokin MIP-1α hauptsächlich das Signal.
