Zusammenfassung
Das Ende der 60iger und die 70iger Jahre werden als Jahre des großen Fortschritts auf dem Gebiet der Hepatitis-B-Forschung in die Geschichte der Virologie eingehen. 1965 wurde das sog. „Australia-Antigen“ durch Blumberg entdeckt [4], und 1968 konnte dieses Antigen durch Prince mit der Hepatitis B assoziiert werden [18]. Bei dem Antigen, das erstmals im Serum eines australischen Ureinwohners mit Hilfe von Antikörpern, die im Serum von Patienten mit häufigen Transfusionen vorhanden waren, gezeigt werden konnte, handelt es sich, wie wir heute wissen, um das Oberflächenantigen (HBsAg) des Hepatitis-B-Virus (HBV). Es folgte 1970 die Entdeckung des HBV, eines 42 nm messenden, aus Kern und Hülle bestehenden Viruspartikel, das auch als Dane-Partikel bezeichnet wird [5]. 1971 wurde erstmals das Antigen des Viruskerns (HBcAg) [2] und 1972 erstmals das sog. HepatitisB-e-Antigen (HBeAg) [13], dessen Lokalisation im Dane-Partikel noch nicht eindeutig geklärt ist, beschrieben. Antikörper gegen diese Antigene werden Anti-HBs, Anti-HBe und Anti-HBc genannt. In den folgenden Jahren konnte eine zirkuläre doppelsträngige DNA und eine DNA-Polymerase im Viruspartikel gezeigt werden [21]. Durch Klonierung der Nukleinsäure im bakteriellen System konnten weite Teile der Nukleinsäuresequenz des Gens und damit auch der Aminosäuresequenz der viralen Proteine aufgeklärt werden [29]. Die Prävention der Hepatitis B ist durch passive Immunisierung mit hochtitrig Anti-HBs enthaltendem Immunserumglobulin und durch aktive Immunisierung mit einer HBsAg-enthaltenden Totvakzine möglich geworden [11].
