Zusammenfassung
Peptide aus den Herzvorhöfen verschiedener Spezies (atriales natriuretisches Peptid = ANP, Atriopeptin, Cardiodilatin) können die Natriurese stimulieren und isolierte Gefäßmuskulatur zur Erschlaffung bringen (1, 2). Die zellulären Mechanismen für diese Effekte des ANP sind bisher nicht vollständig bekannt. Es ist jedoch wahrscheinlich, daß zumindest am Gefäßmuskel die Stimulation einer membranständigen Guanylatcyclase und eine daraus resultierende Steigerung des zytosolischen cGMP (4) einen Zwischenschritt auf dem Weg zur Gefäßerschlaffung darstellt. Die andere große Substanzgruppe mit vasodilatatorischer Wirkung, die nach der herrschenden Auffassung über eine Erhöhung des zellulären cGMP (als Folge einer Stimulation zytosolischer löslicher Guanylatcyclase durch Zwischenprodukte, z.B. S-Nitrosothiole) die Gefäßmuskulatur zur Erschlaffung bringt, sind die „Nitrate“ wie Nitroglycerin, Isosorbiddinitrat und Natriumnitroprussid (7). Es besteht also die Möglichkeit, daß die auf die Bildung von zytosolischem cGMP folgenden Schritte, welche zur Relaxation der Gefäßmuskelzelle führen, für ANP und Vasodilatatoren vom Nitrattyp identisch oder zumindest ähnlich sind.
