Abstract
We have recently developed methods for the quantitative measurement of B-glucuronidase (GUS) reporter gene activity of transformants of Neotyphodium endophytes during their growth in perennial ryegrass (2). When transformants express the reporter gene constitutively, GUS activity can be used as an indicator of the distribution of in planta endophyte metabolic activity (2). Because the concentration of endophyte metabolic activity in grass tissue is a result of both the concentration of endophyte hyphae and their metabolic state, we have now combined GUS assays with methods for the determination of biovolume of endophyte hyphae in plant sections to assess the metabolic state of the endophyte mycelium.
