Role of DNA Repair in Mutation and Cancer Production

  • Chapter

Abstract

The somatic mutation theory of carcinogenesis was postulated long before there was knowledge of the molecular structure of genetic material, the physical, chemical and/or biological nature of carcinogens, or the nature of the interaction between cellular DNA and the carcinogenic agents (Boveri, 1929). The concept was originally introduced to account for two of the many observations of neoplastic growth; (a) the unlimited variety of tumor types, and (b) the fact that, after cell division, the daughter cells maintain their neoplastic properties.
