Abstract
We report that extensive variable (V) gene diversity is generated in the Balb/c memory response to phosphocholine-keyhole limpet hemocyanin (PC-KLH). Antibodies found in the primary response to PC-KLH (referred to as Group I) express the VH1 gene, are T15 idiotype positive and bind the haptens PC and nitrophenyl PC (NPPC). In contrast, the memory response exhibits new antibodies (Group II) that bind appreciably only NPPC, are T15 idiotype negative and express different VH and VL genes.
VH sequence analysis of a prototype Group II antibody indicates that the M141 VH gene is utilized by this memory-derived hybridoma. This VH gene in association with the Vk1-3 light chain (L) generates a Group II binding site. The M141 VH gene is also used by four Group II hybridomas that express the lambda-1 (λ1) L chain. Northern and Southern blot analyses indicate that VH gene members of the Q52(M141) and J558 VH families can be used with non-Group I L chains to generate Group II antibodies during immune maturation to PC-KLH. Our findings indicate that the establishment of immune memory in the anti-PC-KLH response is accompanied by the recruitment of B cells using novel VH and VL genes in new associations.
