Abstract

Platelet-activating factor [PAF (1-O-alkyl-2-acetyl-sn-glycero-3-phosphocholine); also called AGEPC or PAF acether] is an ether phospholipid implicated in several human pathologies including asthma and shock, cardiac and systemic anaphylaxis, immune and renal disorders, and a variety of inflammatory conditions (Braquet et al., 1987; Prescott et al., 1990). In addition, several lines of evidence suggest the implication of PAF in various pathophysiologic processes of the central nervous system (CNS). Indeed, several PAF receptor antagonists have been used successfully to limit brain damage in animal models of ischemia (Panetta et al., 1987; Spinnewyn et al., 1987; Birkle et al., 1988; Kochanek et al., 1987; Szabo et al., 1991), stroke (Lindsberg et al., 1990), and experimental allergic encephalomyelitis (Howat et al., 1989). Recent evidence shows a role for PAF in the release of excitatory amino acid neurotransmitters (Clark et al., 1991) and in long-term potentiation (Del Cerro et al., 1990).
