A GC–MS-SIM Simultaneous Determination of Ligustilide and Butylidenephthalide from Ligusticum chuanxiong Using SFE

Abstract

A simple capillary GC–MS method for the simultaneous determination of ligustilide (LGT) and butylidenephthalide (BDP) in Ligusticum chuanxiong is described. LGT and BDP were obtained by supercritical fluid extraction (SFE), and muscone was used in this method as an internal standard for quantification. MS detection was performed in SIM mode to increase the sensitivity. The tested ranges were 20–1,000 μg mL−1 for LGT and 2–100 μg mL−1 for BDP. This method was fully validated and showed good performances in terms of recovery (98.2–101.5%) and precision (1.21–3.52%). Finally, the method was applied to the analysis of LGT and BDP in L. chuanxiong by SFE.
