High Performance Liquid Chromatographic Determination of Esculetin and Daphnetin in Urine and Plasma

Summary

We have developed and validated a rapid quantitative chromatographic technique and solvent extraction protocol for the quantitation of esculetin and daphnetin in plasma and urine. The linear range was 0.5–100 μg mL−1. The % recovery for both compounds was found to be in excess of 70%. Intra- and inter-assay precision and accuracy was determined for esculetin and daphnetin following extraction from both plasma and urine, and found to be excellent. It is intended that this method will facilitate the analysis of biological samples obtained from in vivo clinical trials, thereby expediting the pharmacological evaluation of these compounds.
