Résumé:
p53 est une protéine impliquée dans la réparation de l’ADN ou l’induction de l’apoptose après un stress cellulaire. C’est l’un des marqueurs biologiques les plus étudiés en cancérologie; malgré le nombre de publications sur p53, le niveau de preuve semble insuffisant pour l’utiliser en clinique comme facteur prédictif de réponse ou comme facteur pronostique.
De nouvelles thérapeutiques cependant sont testées; en particulier il peut s’agir de thérapeutiques vaccinales ou de thérapies géniques en cours d’expérimentation.
Abstract:
p53 is an important protein in repairing cell DNA or triggering apoptosis after an injury. It is one of the most investigated molecular markers in cancer; many papers have been published about this biomarker. Nevertheless, the level of evidence for the use of p53 to predict recurrence, progression and mortality is not clear because of the heterogeneity of the data.
As a predictive marker for treatment response, p53 should be considered with caution for the same reasons.
New treatments are being tested. It will be examined whether vaccination p53 peptide can boost an immune response to breast or ovarian cancer and what the side effects are of the vaccine or p53 gene therapy using an adenovirus vector or administration of the Oncolytic Adenovirus ONYX-015.
