Optimal mode of operation for temperature induction of bacteriophage λQ mutant in Escherichia coli

Abstract

An epidemic model is presented to provide theoretical considerations for temperature induction of bacteriophage λ in Escherichia coli. Based upon model predictions for bioreactor operations, the optimal mode for reporter-protein induction is fed-batch since augmentation of susceptible cells enhances λ infection in batch cultures and washout of infected cells decreases induction performance in CSTR operations. Experimental results also support that batch mode of operation is better than CSTR.
