Summary
10 and 20 mg of N-Propyl-ajmalin-hydrogentartrate (N-PAB, Neo-Gilurytmal®) were administered i.v. and orally respectively to healthy volunteers. In the study14C-labelled N-PAB was used. The pharmacokinetics and the metabolic behaviour was examined. A fast and complete absorption of the compound could be observed. The bioavailability was 78%. The terminal plasma elimination half-life (β-phase) was in the range of 4 to 6 hours. A total of 33% of the administered radioactivity was excreted via the kidney. 35 to 48 % of the radioactivity found in urine was unchanged N-PAB. The excretion-kinetics of the three main metabolites as well as of the parent compound were determined. The possible presence of non-metabolizers is suggested.
Zusammenfassung
Nach Applikation von N-Propyl-ajmalin-hydrogentartrat (N-PAB, Neo-Gilurytmal®) in den Dosierungen 10 mg i.v. und 20 mg p.o. wurden Pharmakokinetik und Metabolismus an sechs gesunden männlichen Freiwilligen untersucht. Für die Untersuchung wurde14C-markiertes N-PAB verwendet. N-PAB wird schnell und vollständig resorbiert. Die Bioverfügbarkeit beträgt 78%. Die Eliminationshalbwertszeit für die Muttersubstanz liegt zwischen 4 und 6 Stunden. Die Elimination der14C-Radioaktivität erfolgt zu 33% über die Niere. Der Anteil der Originalsubstanz an den Ausscheidungsprodukten beträgt 35 —48%. Die Ausscheidungskinetik der 3 Hauptmetaboliten, die deutlich langsamer als N-PAB eliminiert werden, wurde bestimmt. Auf die Möglichkeit des Auftretens von Nichtmetabolisierern wird hingewiesen.
