Experience in protein-bound iodine estimation by a modified alkaline ashing procedure

(1) The Foss, Hankes and Van Slyke2 method for determining PBI in serum has been modified to reduce cross-contamination of analyticals, occurrence of amorphous carbon, and falsely high PBI values.
(2) The range found for 85 normal individuals was 3.3 to 6.9 µg iodine/100 ml. serum. A range of 0.1 to 3.1 was obtained in 19 suspected hypothyroid cases. Tests on 48 suspected hyperthyroid cases gave a range of 5.2 to 19.2 with only two values falling below 7.2 µg. iodine/100 ml. of serum.
(3) Results obtained are discussed and compared to those of other authors.
(4) Factors unrelated to abnormalities of the thyroid gland but which affect the PBI level are summarised.
(5) This method of PBI estimation appears to be reliable and suitable for research purposes. For PBI determinations on a large routine scale an autoanalyser is advisable.
