Zusammenfassung
Ferrocen ist eine eisenorganische Verbindung von Sandwich-Charakter mit Redoxeigenschaften. Es eignet sich als Katalysator, z.B. als Additiv in Treib- und Schmierstoffen; auch als Medikament hat es zur Lieferung von Eisen bei Anämieerkrankungen gedient. Der Stoff wird über Magen-Darm und Lunge rasch resorbiert und spaltet im Organismus Eisen ab. Auf dem freigesetzten Eisen beruhen die beobachteten akut-toxischen Wirkungen: Gewebsreizung bis zur Zellnekrose; gespeichert wird Eisen, nicht aber Ferrocen selbst. Ferrocen wird im Stoffwechsel hydroxyliert und nach Kopplung mit Glukuronsäure rasch über die Nieren ausgeschieden. Die akute und subakute Toxizität ist gering. Bei Inhalation im Tierversuch wird das Riechepithel konzentrationsabhängig geschädigt. Im Langzeitversuch (6 Monate) werden an Hunden ab 100 mg/kg/Tag oral Vergiftungszeichen in Form von verstärkter Blutbildung, Leberschädigung und Hämosiderinablagerungen in allen parenchymatösen Organen gefunden. Mutagenitätsuntersuchungen in einer Reihe von mikrobiellen und Warmblüterzellsystemen brachten negative Ergebnisse; lediglich ein Versuch an Drosophila brachte ein zweifelhaftes Ergebnis. Im Kanzerogenitätstest nach intramuskulärer Applikation wurden lokale Tumoren beobachtet, wie sie von einer Reihe anderer, eisenliefernder Verbindungen bekannt sind.
