Summary
The effect of thymopentin on the mortality rate of mice treated with lethal doses (LD90) of herpes virus 2 and on the cytotoxic T cell activity after sublethal doses (LD10) of herpes virus was investigated in two series of experiments. Doses of 1, 0.1 or 0.01 ng of thymopentin per g/mouse were administered i.p. in each experiment, either 3 days before, 3 days (66 h) after, or 3 and 6 days after the herpes virus infection. The cumulative mortality rate was evaluated 10 days after the infection. Cytotoxic T cell activity was measured 3, 7 and 14 days after the infection. The 0.1-ng dose of thymopentin reduced the mortality rate to less than 50% (p = 0.0000) if it was administered 3 days before the infection. A single injection of any dose after infection did not reduce the mortality at all, while two injections of 0.1 ng reduced it by about 25% (p = 0.0038). A 1-ng dose showed a mild but significant reduction (p = 0.0313) if it was applied 3 days before the infection. The cytotoxic T cell activity was either not influenced or significantly modified (p < 0.05), i.e. increased or decreased as compared to the control, depending on the dose and timing of thymopentin. A correlation between increased cytotoxic T cell activity and protection against mortality can be demonstrated, while no protection was observed in dose regimens where the cytotoxic T cell activity became reduced. The results are discussed in connection with earlier clinical studies in which the beneficial effect of thymopentin has been demonstrated in frequently relapsing herpes labialis and herpes genitalis patients.

Dans deux séries d’expériences, on a étudié l’effet exercé par la thymopentine sur le taux de mortalité de souris traitées par des doses léthales (DL90) de virus herpétique 2 ainsi que sur l’activité cytotoxique des cellules T après des doses subléthales (LD10) de virus herpétique. Une dose de 1 ng, de 0.1 ng ou de 0.01 ng de thymopentine a été administrée par voie intrapéri-tonéale à la souris au cours de chaque expérience, 3 jours avant, 3 jours après (66 h) ou 3 et 6 jours après l’infection par le virus herpétique. Le taux cumulatif de mortalité a été évalué 10 jours après l’infection. L’activité cytotoxique des cellules T a été mesurée 3, 7 et 14 jours après l’infection. La dose de 0.1 ng de thymopentine a fait baisser le taux de mortalité de plus de 50% (p = 0,0000) lorsqu’elle était administrée 3 jours avant l’infection. Une injection unique après l’infection, quelle que soit la dose, n’a pas du tout fait baisser la mortalité, alors que deux injections de 0.1 ng l’ont diminuée de 25% environ (p = 0,0038). La dose de 1 ng déterminait une réduction mineure mais significative (p = 0,0313) lorsqu’elle était administrée 3 jours avant l’infection. L’activité cytotoxique des cellules T était soit inchangée, soit significativement modifiée (p < 0.05), c’est-à-dire accrue ou diminuée en comparaison des témoins, selon la dose de thymopentine et le moment de l’administration. On peut démontrer l’existence d’une corrélation entre l’augmentation de l’activité cytotoxique des cellules T et la protection contre la mortalité, alors qu’aux schémas posologiques s’accompagnant d’une réduction de l’activité cytotoxique des cellules T, aucun effet protecteur néa été observé. Ces résultats sont discutés à propos d’études cliniques antérieures, dans lesquelles l’effet bénéfique de la thymopentine a été démontré chez des patients souffrant d’herpès labial et génital à récidives fréquentes.

Die Wirkung von Thymopentin auf die Mortalitätsrate von Mäusen, die mit einer tödlichen Dosis (LD90) von Herpesvirus 2 behandelt worden waren, sowie auf die zytotoxische T-Zell-Aktivität nach subletalen Dosen (LD10) von Herpesvirus wurde in zwei Versuchsreihen geprüft. Thymopentindosen von 1, 0.1 oder 0.01 ng/g/Maus wurden in jedem Versuch i.p. verabreicht, entweder 3 Tage vor, 3 Tage (66 h) nach, oder 3 und 6 Tage nach der Infektion mit Herpesvirus. Die kumulative Mortalitätsrate wurde 10 Tage nach der Infektion analysiert. Die zytotoxische T-Zell-Aktivität wurde 3, 7 und 14 Tage nach der Infektion gemessen. Eine Dosis von 0.1 ng Thymopentin reduzierte die Sterblichkeitsrate auf weniger als 50% (p =0.0000), sofern das Medikament 3 Tage vor der Infektion verabreicht wurde. Eine Einzelinjektion einer beliebigen Dosis nach der Infektion verringerte die Sterblichkeit überhaupt nicht, während zwei Injektionen zu 0.1 ng die Mortalität um zirka 25 % reduzierten (p = 0.0038). Zu leichter, aber signifikanter Reduktion kam es mit 1 ng (p = 0.0313), wenn diese Dosis 3 Tage vor der Infektion verabreicht wurde. Die zytotoxische Zellaktivität blieb entweder unbeeinflusst oder wurde signifikant modifiziert (p < 0.05), d.h. im Vergleich zur Kontrollgrupe je nach dem Dosis/Zeit-Verhältnis von Thymopentin erhöht oder verringert. Eine Korrelation zwischen der erhöhten zytotoxischen T-Zell-Aktivität und dem Schutz gegen Sterblichkeit war nachweisbar, während mit Dosen, welche die zytotoxische T-Zell-Aktivität verringerten, kein Schutz gegen Mortalität ersichtlich war. Die Resultate werden im Zusammenhang mit früheren klinischen Studien diskutiert, in welchen der günstige Effekt von Thymopentin bei häufigen Rückfallen von Herpes labialis und Herpes genitalis nachgewiesen wurde.

In due serie d’esperienze si è studiato l’effetto esercitato dalla thymopentine sul tasso di mortalità di topi trattati con dosi letali (DL90) di virus erpetico 2, corne pure l’azione di questo farmaco sull’attività citotossica délie cellule T dopo la somministrazione di dosi subletali (LD10) di virus erpetico. Una dose di 1, 0,1 o di 0,01 ng di thymopentine venne somministrata per via i.p. al topo nel corso di ogni esperienza, vale a dire 3 giorni prima, 3 giorni dopo (66 ore) oppure 3 e 6 giomi dopo l’infezione col virus erpetico. Il tasso cumulativo di mortalità è stato evaluato 10 giorni dopo l’infezione. L’attività citotossica delie cellule T venne misurata 3, 7 e 14 giorni dopo i’infezione. Un dosaggio di 0,1 ng di thymopentine ha provocato l’abbassamento del tasso di mortalità di più di 50% (p = 0,0000) allorché la dose era stata somministrata 3 giorni prima dell’infezione. Un’iniezione unica dopo l’infezione, in qualunque dosaggio, non ha fatto per nulla diminuire la mortalità; per contro, due iniezioni di 0,1 ng l’hanno abbassata circa del 25 % (p = 0,0038). La somministrazione di una dose di 1 ng provocava una riduzione minore ma significativa (p = 0,0313) allorché veniva somministrata 3 giorni prima dell’infezione. L’attività citotossica delie cellule T era, sia inalterata, sia modificata in modo significativo (p < 0,05), vale a dire aumentata o diminuita per rapporto ai controlli, secondo la dose di thymopentine e il momento della somministrazione. Si può dimostrare l’esistenza d’una correlazione fra l’aumento dell’attività citotossica delie cellule T e la protezione contro la mortalità, mentre negli schemi di posologia avenu corne conseguenza una riduzione dell’attività citotossica delie cellule T, non si osservò alcun effetto protettore. Questi risultati sono stati discussi a proposito di studi clinici precedenti, nel corso dei quali è stato dimostrato l’effetto benefico delia thymopentine in pazienti affetti da herpes labiale e genitale caratterizzati da recidive frequenti.

Se investigó el efecto de la timopentina sobre la tasa de mortalidad de ratones tratados con dosis letales (DL90) de virus del herpes tipo 2 y sobre la actividad de las células T citotóxicas tras dosis subletales (DL10) de virus de herpes en dos series de experimentos. Se administró 1, 0,1 o 0,01 ng de timopentina por ratón i.p. en cada experimento, ya sea 3 dias antes, 3 días (66 h) después o 3 y 6 días después de la infección por virus del herpes. La tasa de mortalidad acumulativa se evaluó 10 dias después de la infección. Se midió la actividad de las células T citotoxicas 3, 7 y 14 dias después de la infección. 0,1 ng de timopentina redujo la tasa de mortalidad en más del 50% (p = 0,0000) caso de administrarse 3 días antes de la infección. Una inyección única de cualquier dosis después de la infección no redujo lar mortalidad en absoluto, mientras que dos inyecciónes de 0,1 ng la redujeron en un 25% aprox. (p = 0,0038). 1 ng presentó una ligera pero significativa reducción (p = 0,0313) caso de aplicarse 3 días antes de la infección. La actividad de las células T citotoxicas no se vio influida ni modificada significativamente (p < 0,05), es decir, aumentada o disminuida en comparación con el control, según la dosis y tiempo de la timopentina. Se puede demostrar una correlación entre el aumento de la actividad de las células T citotóxicas y la protección contra la mortalidad, mientras que no se observó protección alguna en regimenes posológicos en donde la actividad de las células T citotóxicas se redujo.
