Summary
After intravenous injections of hexadimethrine-bromide (Polybrene® Abbott) into rats, we were able to show histological and histochemical disturbances in the kidneys and a severe progressive oliguria, azotemia and altered excretion of electrolytes. Histologically, the changes were characterized by an almost selective necrosis of the thick ascending segment of Henle’s loop and a presumably secondary focal degeneration of the cortical portion of the nephron. Histochemically, the activity of succinic dehydrogenase, of alkaline and acid phosphatases, and of non-specific esterase disappeared in the necrotic Henle’s loops, and decreased greatly in the foci of degeneration. In the histologically normal portions of the cortex the activities of the acid phosphatase and non-specific esterase were higher than in the controls. The activity of the other enzymes remained normal.
Zusammenfassung
Die Verfasser haben nach intravenöser Verabreichung von 5 mg Hexadimethrin-bromid (Polybrene®, Abbott) die Nierenfunktion von Ratten sowie die in diesem Organ auftretenden histologischen und histochemischen Abweichungen untersucht und kommen zu der Feststellung, daß nach der Behandlung eine hochgradige und anhaltende Oligurie, Azotämie und Störung in der Elektrolytentleerung auftritt.
Histologisch kennzeichnend ist die fast immer selektive Nekrose des dicken aufsteigenden Teiles der Henleschen Schleifen und die vermutlich sekundäre, herdförmige Degeneration des Rindenanteils des Nephron. Histochemisch hört die Aktivität der untersuchten Enzyme (Succindehydrogenase, alkalische und saure Phosphatase, nichtspezifische Esterase) in den nekrotischen Henleschen Schleifen auf und läßt in den lädierten Herden stark nach. In den histologisch normalen Anteilen der Rinde ist die Aktivität der sauren Phosphatase und der nichtspezifischen Esterase höher als bei den Kontrollen und die der anderen drei Fermente normal.
