Abstract
Chromosome studies on peripheral blood cultures were performed in 18 children receiving Periactin® for therapeutic reasons. The average frequency of aberrations after therapy was somewhat higher than before therapy. The differences were not significant. (P>0.05). Thus, results did not give a distinct suggestion of Periactin®-induced cytogenetic damage in the patients tested.
Because of positive results in previous in vitro tests with human lymphocytes further studies in a greater number of patients with a larger dosage scale appear necessary. In addition, further experimental mutagen research with in vivo systems may help to clarify mutagenic activity of Cyproheptadine.
Zusammenfassung
Von 18 Kindern, die aus klinischer Indikation mit Nuran® (1 bis 8 mg/die) behandelt worden waren, wurden Chromosomenuntersuchungen durchgeführt. Die Aberrationshäufigkeit lag nach der Therapie im Durchschnitt etwas höher als vor Therapiebeginn. Eine generelle statistische Sicherung dieser Abweichung war jedoch auf dem 5%-Niveau nicht möglich. Lediglich drei Einzelwerte (zwei davon nach zweitägiger Therapie) lagen signifikant höher als die zugehörigen Kontrollwerte. Die Ergebnisse brachten damit keinen eindeutigen Hinweis auf eine cytogenetisch schädliche Wirkung des Nuran im untersuchten Patientenkollektiv.
