Summary
A method for lactate determination is described which is based upon oxidation to pyruvate catalyzed by l-lactic dehydrogenase (LDH) from baker's yeast. Methylene blue acts as an electron acceptor and is reduced to its colourless leuco form. The leuco dye is oxidized by oxygen present in the solution producing a reversible system. Reproducible signals are obtained if the solution is saturated with oxygen. The measuring range depends on the concentration of methylene blue and has to be adapted to the measuring problem. Lactate determinations in real samples by the new method agreed with determinations made by the classical NADH method.
