Preparation of lyophilised drug spiked calf's serum

In this study, a survey is given of the preparation of lyophilised antiepileptic quality control serum specimens, as used in the joint international test on antiepileptic drugs (Dijkhuiset al. 1979).
Calfs serum, previously filtered through cellulose asbestos pads was used as a matrix and spiked with 8 antiepileptic drugs; the drug concentrations were calculated on basis of weighed-in values, corrected for increase of volume after reconstitution.
Difficulties, encountered by components, eluted from rubber andPvc tubes and stoppers during filtration and lyophilisation procedures are discussed.
As an example, the preparation of a batch of lyophilised quality control serum is described in detail.
A preliminary report is given on serum filtration using the Pellicon Cassette Ultrafiltration System, developed by Millipore. (Pharm. Weekblad Sci. Ed.1, 185-196)
