Summary

2/3 of the Evipal degraded by incubated rat liver slices are transformed in 2 unknown barbiturates. The structure of these products is discussed.

Liver slices of male albino rats destroy about 70% of the Evipal, those of females only 30–40%. Under the same conditions 10% of Thiopental is degraded, no sex difference is noted. Correspondingly after administration of Evipal female rats “sleep” 4 times longer than males. But after injection of Thiopental the duration of anaesthesia in male and female rats is not different.

Likewise homogenate and supernatant of rabbit liver destroy Evipal with a much higher speed than Thiopental.

The in vitro degradation of Evipal can be inhibited by the injection of 50 mg/kg. i.p. of β-diethylaminoethyl-phenyldiallylacetate 1 hour before decapitation of the rats.

As discussed the biotransformation of Evipal, but not of Thiopental is delayed by this aceticacid ester and enhanced by the male sex hormone.
